Significance of measurement:
NR (EC 1.7.1.3) is widely found in plants, it is the key enzyme for the conversion of plant nitrate nitrogen to ammoniacal nitrogen, and also an inducible enzyme, which has an impact on crop yield and quality.
Principle of Determination:
NR catalyses the reduction of nitrate to nitrite, NO3ˉ +NADH+H+→ NO2ˉ +NAD+ +H 2 O; the generated nitrite can be quantitatively generated with p-aminobenzenesulphonic acid and α-naphthylamine to produce red azo compounds in acidic conditions; the generated red azo compounds have the maximum absorption peak at 540 nm, which can be measured by spectrophotometric method.
The red azo compounds can be determined spectrophotometrically:
Visible spectrophotometer/enzymometer, water bath, bench-top centrifuge, adjustable pipette, micro-quartz cuvette/96-well plate, mortar and pestle, ice and distilled water.
