Measurement significance:
NR (EC 1.7.1.3) is widely found in plants and is a key enzyme in the conversion of plant nitrate nitrogen to ammoniacal nitrogen, as well as an inducible enzyme, which has an impact on crop yield and quality.
Principle of Determination:
NR catalyses the reduction of nitrate to nitrite, NO3ˉ +NADH+H+→ NO2ˉ +NAD+ +H 2 O; the generated nitrite can be quantitatively generated with p-aminobenzenesulphonic acid and α-naphthylamine to produce red azo compounds in acidic conditions; the generated red azo compounds have the maximum absorption peak at 540 nm, which can be measured by spectrophotometric method.
Apparatus and supplies to be provided:
Visible spectrophotometer, water bath, benchtop centrifuge, 1 mL glass cuvette, mortar and pestle, ice and distilled water.
