Measurement significance:
FAS is a key enzyme in fatty acid synthesis, catalysing the production of long-chain fatty acids from acetyl coenzyme A and malonyl coenzyme A. FAS is commonly expressed in various tissues and cells, and is abundantly expressed in mammalian liver, kidney, brain, lung and mammary gland, as well as adipose tissue.
Principle of Assay:
FAS catalyses the production of long-chain fatty acids and NADP+ by acetyl CoA, malonyl CoA and NADPH; NADPH has an absorption peak at 340 nm, while NADP+ does not; the rate of decrease in light absorption at 340 nm was measured to calculate FAS activity.
