Name：Citrate Synthase(CS)Assay Kit

Determination method: Spectrophotometric method

Specification: 25 tubes/12 samples

Storage condition: low temperature and keep away from light

Validity: 6 months

Note: 2-3 samples with large expected differences must be taken for pre-calibration before formal determination.

Measurement significance:

CS (EC 2.3.3.1) is widely found in the mitochondrial matrix of animals, plants, microorganisms and cultured cells, it is the first rate-limiting enzyme of the tricarboxylic acid cycle, and is one of the main regulatory sites of the tricarboxylic acid cycle.

Principle of determination:

CS catalyzes the production of citrullinated coenzyme A from acetyl CoA and oxaloacetate, which is further hydrolysed to produce citric acid; this reaction leads to the conversion of colourless DTNB to yellow TNB, with characteristic absorbance value at 412nm.

Apparatus and supplies to be provided:

Visible spectrophotometer, benchtop centrifuge, water bath, adjustable pipette, 1mL glass cuvette, mortar and pestle, ice, anhydrous ethanol and distilled water
