Measurement significance:
β-GAL (EC 3.2.1.23) is widely found in animals, plants, microorganisms and cultured cells, and is able to catalyse the hydrolysis of the β-galactoside bond in β-galactoside compounds, in addition to its transgalactosidic effect. β-GAL not only releases stored energy for rapid plant growth, but also catalyzes the hydrolysis of polysaccharides, glycoproteins, and glycoproteins in normal polysaccharide metabolism, metabolism of cell wall components, and degradation of the cell wall during senescence. It also catalyses the hydrolysis of polysaccharides, glycoproteins and galactose residues at the end of galactolipids, releasing free galactose.
Principle of Determination:
β-GAL decomposes p-nitrophenyl-β-D-galactopyranoside to produce p-nitrophenol, which has a maximum absorption peak at 400 nm, and β-GAL activity is calculated by measuring the rate of increase in absorbance value.

Visible spectrophotometer, benchtop centrifuge, water bath, adjustable pipette, 1 mL glass cuvette, mortar and pestle, ice and distilled water.
