Significance of the assay:
β-GAL (EC 3.2.1.23) is widely found in animals, plants, microorganisms and cultured cells, and is able to catalyse the hydrolysis of the β-galactoside bond in β-galactoside compounds, in addition to its transgalactosylation. β-GAL not only releases stored energy for rapid growth of plants, but also catalyses the hydrolysis of polysaccharides, glycoproteins, and cell walls during normal polysaccharide metabolism, metabolism of cell wall components, and degradation of the cell wall during senescence. It also catalyses the hydrolysis of polysaccharides, glycoproteins and galactose residues at the end of galactolipids, releasing free galactose.
Principle of Determination:
β-GAL decomposes p-nitrophenyl-β-D-galactopyranoside to produce p-nitrophenol, which has a maximum absorption peak at 400 nm, and β-GAL activity is calculated by measuring the rate of increase in absorbance value.
