Measurement significance:
LDH (EC 1.1.1.27) is widely found in animals, plants, microorganisms and cultured cells and is a terminal enzyme of the glycolytic pathway, catalysing the reversible reaction between pyruvate and lactate, accompanied by interchanges between NAD+/NADH.
Principle of measurement:
LDH catalyses the oxidation of lactate by NAD+ to form pyruvate, which further interacts with 2, 4 - dinitrophenylhydrazine to form dinitrophenylhydrazone pyruvate, which appears brownish red in alkaline solution, with the depth of colour being proportional to the concentration of pyruvate.

Visible spectrophotometer, thermostatic water bath, benchtop centrifuge, adjustable pipette, 1 mL glass cuvette, mortar and pestle, ice and distilled water.
